Mitochondrial respiration

The Oroboros Oxygraph-2k (O2k) evaluates mitochondrial function and efficiency, i.e. respiration capacity, in mitochondrial preparations and living cells.

Applications in brief

Upon sample loading, predefined or adjusted Substrate-Uncoupler-Inhibitor-Titration (SUIT) protocols are applied in which substrates, inhibitors, and uncoupler, are titrated in a sequence to stimulate or inhibit the enzymes and complexes of the electron transfer pathways. This facilitates evaluation of different coupling and pathway control states and provides detailed information on function of the electron transfer pathway components and the phosphorylation system.

Which samples may I use?

Fresh tissue is recommended, although frozen tissue may be used upon thorough protocol optimization

  • living and permeabilized cells (fresh or cryopreserved)
  • permeabilized muscle fibers

Location

The O2k equipment is located on the 4th floor in the Skou building. Therefore, thoroughly evaluate your experiment steps (e.g. equipment preparation, sample preparation, centrifugation) prior to experimental onset.
