MSI - Mass Spectrometry Imaging

TimsTOF fleX MALDI-2 instrument Source: Bruker Daltonics

The timsTOF fleX MALDI-2 is a high-performance mass spectrometer that integrates Trapped Ion Mobility Spectrometry (TIMS) with Matrix-Assisted Laser Desorption/Ionization (MALDI) imaging. The MALDI-2 post-ionization dramatically enhances sensitivity for a comprehensive, untargeted spatial view of biomolecules directly from tissue.

Key Features:

  • Technology: Uses MALDI with MALDI-2 post-ionization to greatly boost signal, particularly for lipids and metabolites.
  • 4D Resolution: Delivers high spatial resolution (~5-10 µm) paired with ion mobility separation, adding a fourth dimension (x, y, m/z, ion mobility) for superior molecular specificity.
  • Sample Compatibility: Supports both FFPE and fresh-frozen tissues with dedicated protocols. Please contact us for more detailed discussion on sample prep protocols.
  • Untargeted Discovery: Enables hypothesis-free imaging of thousands of molecules without pre-defined panels.
  • Multimodal: The "FLEX" design allows seamless switching between MALDI and Electro-spray ionization (ESI) ion sources on a single instrument.

Multiomic Applications:

  • Spatial Metabolomics & Lipidomics: Direct imaging of endogenous small molecules and lipids.
  • Spatial Proteomics: Peptide and protein imaging from FFPE tissues via on-tissue digestion.
  • Spatial Glycomics: Mapping of N-linked glycans.

Data Output: Generates high-dimensional datasets for visualization and analysis in specialized software, (e.g. SCiLS Lab) or more customed data analysis pipelines.
